I went with the Diablo tuner and Diablew tune route. It definitely helped! I run heavy tires and tow frequently. If I remove the tune the truck becomes a super slug - no fun to drive!
Shop diagnosed today and says it's definitely ring/pinion wear and needs to be replaced.

Serviced the rear diff and didn't notice anything abnormal. Refilled with RP 75W-90. Still have the noise. I'm getting the tires rotated and balanced today just to rule that out.
The sound is most noticeable taking off from a stop up to about 40mph. It's subtle, but the pitch and frequency change with increased RPM/speed. If I'm just cruising or at freeway speed I don't really notice it. Sounds like it's coming from the rear end/driveline area. Trans seems to shift...

With the key on (no start) I only get a reading on the M1 trans temp. Maybe the others will work when actually started, I'll check next time I drive it.
Yea I watched both parts of that video from South Main Auto a while ago. It was very informative. I'm going to need to dig in a bit deeper. I think the key will be finding the fuse with the 1.1amp draw. There are fuses that show different amp draws, but I haven't checked all of them yet.
Ok here's my build. I labeled them M1, M2, and M3. I'm not sure if I have the Pro version of Torque, but I know I paid for it a while back. I'll give it a spin soon.

Good idea. In this case I've been wanting to upgrade my cluster anyway so I think I'll go that route. Even if it doesn't fix the problem I'll get the trans temp gauge I've always wanted and some nicer customized gauges.
I've done some testing and plan on replacing my cluster soon, though I'm not sure if it's the whole problem or not. What I've found is that when I'm measuring a draw of about ~1.1 amps at the negative battery cable, I then do a drop voltage test of the IPC/DIC fuse which shows about 300...
