Could be spark knock. Something is setting off the knock sensors and the ECM is pulling out timing. Try putting a tank of premium in it and see if it changes.

Based on my limited knowledge of the VLOM I would replace it and the gasket or go to an older style LS3 valley cover. Did you replace all lifters when you installed the new CAM? If not that could be a cause of your misfire, I'd go over to LS1Tech and research it.

Make sure you burped all the air out of the system. These things are notoriously hard to get all the air out. You didn't mention if it was over heating or not either.
