I have a Yukon and my father in lay has a 99 suburban. When I turn my AC on in the Yuk the compressor "click's" on. In the sub, no click. He was having problem last year and had a shop recharge it. My question is if the r-134 is low, will that prevent the compressor from clicking on? or could it be a fuse or relay?

The compressor won't kick on without pressure to keep it from burning up. I wrote a how-to on all of that. Check out the tech section.

The burbs take 4 cans. Just monitor your low side pressure if you dont have the manifold gauges to monitor high side pressures.
