I have a 2008 yukon that I use to tow a sailboat(4000lb) 5.3L auto. 110K.
I plan on towing the boat from the East Coast to Puerto Vallarta Mexico. About a 30 hour trip. 18 hours of it across Mexico. My truck has had regular maintenance and no major repairs. What would you guys recommend I have done before the trip and what should I take along with me?

I would service the transmission. I might even go as far as to replace all of the shift solenoids. fuel filter and carry a spare. I might even go as far as to change the fuel pump, your over 100k it be smart money spent. bring a spare fan belt. how new are your tires? brakes? just go over the truck real good to make sure you don't have any fluid leaks that could get bad and strand you. carry enough extra oil and filter and antifreeze.

+1 on this, I have heard a few sketch stories of friends that went to mexico. One of my buddies is going down there soon and decided to fly this time around. Though you can't real fly a boat...

Definitely go over everything, though I was taught to not fix anything prior to a trip in case of the chance of something going wrong with the "fix." I would replace the fuel pump though, GM is notorious for fuel pumps crapping out, we had two go out at 70k and 90k and both were never ran under a quarter tank.

Also, keep an eye on your transmission temp.

Spare battery, alternator, fuel pump, and trans service.

Although the bullet proofing is probably the best advice yet.
