Unfortunately not. You'll need the o-ring. That said, it should only leak if it's parked facing uphill, so there's that. You can just keep an eye on the transmission fluid and add when needed.
The Tech2 will also let you isolate the problem component, if there is one.
My money is on 1/ the trim reset, and then 2/ a loose connector.

Welcome to the forum! Fairly common issue with the 4L80E. There's a weep hole in the yoke that relieves pressure as the driveshaft moves in and out over bumps and such. If the transmission tail shaft o-ring is leaking, on the yoke is where you'll see it. More info here, though it shows a 700R4...
Interesting video. It's hard to separate the camera shake from the TC shake. Also sounds like driveshaft imbalance + bearing runout could be the cause there, but without video of the driveshafts it's hard to say.

Essentially none. You can do things like reset the ALC, run the brake bleed routine, etc, but no settings programming that I know of, and no engine or transmission tuning.
Sounds like a solid plan. Coolant recovery tank leaks are common in that spot. Your hypothesis on the rear air shocks sounds plausible.
Post a pic of the rear end "seep" and we can advise from there.
From what I've read, the valving for the Yellow (4600 HD series) and the Aluminum (5100) is the same but the 5100 series are longer and technically are for lifted trucks.

Circling back on this to answer your question. So far, for my purposes of towing a camper, the 4.10 gears are absolutely perfect. And unloaded, I'm besting the fuel economy on my Denali's by 2-3 MPG in mixed driving. Didn't see that coming.
