I don't know about 4.8 heads. Never done it, but if you want to stay N/A these motors respond very well to cams. It's a heavy truck though so get you one with your focus being on torque as low as possible and a wide curve. A stall and gear will help emmensly. The only downside I see is with the weight of these things if you go too big with the stall your poor 4L60 is gonna be hurting. lol. I do recommend an upgraded Tranny cooler if you stall it as you will be producing a lot more heat through a stall and heat kills transmissions. As for cams and heads I personally would go with the TSP 5.3 PRC stage 2 heads and a 214 or 228R cam. They'll set up the heads the way you want and the combo sells for around $1300. There's guys with LS camaros making well into the 500hp range on those heads. When all else fails you could go with power on a payment plan and spray a little giggle juice. Sorry to ramble so long. Just getting my 2 pennies in.

With that cam and head setup, would I need another cam if I plan on supercharging?

A lot depends on how aggressive you go with the cam. Really good NA cams don't always work well with forced induction.

Juts so you know 4.8 heads are no different than 5.3 heads. There are 2 different casting numbers dependent on what year the head was made. ( Casting Number 706 and 862) but they are both the exact same head used on 4.8's and 5.3's. Both the intake and exhaust valves are 1.89 inches and 1.55 inches and they both have Combustion Chamber Volume of 61.15cc.
